Located 2.5 miles south of the town of Amlwch, the village is close to both Llyn Alaw, the largest body of water on the island, and Parys Mountain, the site of the historic copper mines which lies just to the north. Rhosybol is situated 1½ miles northeast of Penwerthyr.

The ancient church of St Pabo, Llanbabo is dedicated to Saint Pabo: possibly Pabo Post Prydain, one of the leaders among the Britons of the Hen Ogledd following the withdrawal of the Roman legions. Llanbabo is situated 2 miles west of Penwerthyr.
